Various solid supports have been developed in the field of solid-phase peptide synthesis (SPPS) to advance the synthetic efficiency of peptides, oligonucleotides, oligosaccharides and small organic compounds. It is well-known that the swelling property and the solvent compatibility of the resin directly affect the purity and yield of products. To minimize swelling effect and solvent compatibility problems, we designed a new method, engineering core─shell type resin by bi-phasic functionalization. Using this method, we can easily control the thickness of the shell layer and design the core region of the resin. The engineered core─shell-type resin was evaluated by synthesizing the JR 10-mer and ACP(65-74). |
