Copolymerd of N-isopropylacrylamide(NIPAM), methacrylic acid(MAA) and octadecyl acrylate(ODA)(P(NIPAM-co-MAA-co-ODA) were prepared. Since NIPAM segments are hydrophilic and ODA are hydrophobic, the copolymer could be assembled into micelles in an aqueous solution. Glucose oxidases(GODs) were conjugated to MAA residues of the polymeric micelles using 1-ethyl-3-[3′-(dimethylamino)propyl]carbodiimide(EDCI) and N-hydroxysuccinimide(NHS). The GOD-conjugated polymeric micelles were glucose-sensitive in terms of the integrity of micelles. At neutral and alkali conditions, carboxyl groups of MAA are ionized and copolymer chains would take an expanded form. The formation of micelle was somewhat restricted. Glucose is added to the polymer solution, enzymatic reaction of GOD with glucose produces gluconic acid and accordingly the polymer solution become acidic. In this circumstance, carboxyl groups of MAA are unionized and the copolymer could be readily assembled into micelle. |
