The design of nanoparticles mimicking the key features in size, shape and surface molecular organization of biological objects that provide danger-signals are essential in the formulation of effective vaccine. In this taik, I would like to discuss the development of engineered nanoadjuvants that can be used for cancer immunotherapy. Multifaceted tumosomes were synthesized by assembling tumor cell membrane proteins as tumor-associated antigens with two lipid-based adjugants as pathogen characters. We also designed nanocomplexes bearing multivalent immunostimulatory ligands and demonstrated that the multivalent nanocomplexes led to an enhanced stimulation of immune cells and antitumor and immune memory response. Antigen-antibody immune complexes were also developed to directly bind to and cross-link Fc-gamma receptors on DCs, which induced enhanced migration of DCs to draining lymph nodes through the upregulation of the chemokine receptor and cross-presentation against tumor antigen. |
