Isoprenoids are one of the natural product groups that have been considered as attractive molecules owing to the beneficial effects on human health as an antioxidant- and anticancer-agent. In nature, all microbes synthesize isoprenoids from IPP and DMAPP produced by the MVA or the MEP pathway. Although MEP pathway has a potential advantage in carbon yield (MEP pathway consumes 1.255 Glucose for 1 IPP, whereas MVA pathway does 1.5 Glucose ), previous works mainly focused on non-native MVA pathway due to the enigmatic regulation derived MEP pathway in the E. coli. Here, we demonstrated the lycopene production via MEP pathway in E. coli. The native MEP pathway and heterologous lycopene producing pathway were reconstituted based on synthetic biology approach to channel carbon fluxes toward lycopene production, while avoiding possible unknown regulatory system. This system will be applicable to produce valuable isoprenoids as the platform cell factory. |
