제목 Effect of Water-Soluble Additives on Release Profile of BCNU from PLGA Wafer
초록 1,3-bis(2-chloroethyl)-1-nitrosourea(BCNU) is an effective chemotherapeutic agent which has been used clinically for treating malignant glioma. Poly(DL-lactide-co-glycolide)(PLGA) is a well known biodegradable polymer used for drug delivery system as drug carrier. In this study, we investigated the effect of water-soluble excipients such as poly(vinylpyrrolidone)(PVP) or sodium chloride incorporated into BCNU/PLGA on release profiles of BCNU. BCNU loaded PLGA wafers with or without excipients were fabricated by direct compression and In vitro release of BCNU from the wafer was assayed by high performance liquid chromatography (HPLC). Initial release amount and release rate of BCNU increased with increase of PVP or NaCl amount. Molecular weight change, mass loss, morphological change of wafer, and pH of release test media were characterized to investigate the effect of PVP or NaCl on both drug release profile and degradation behavior of wafer.
