제목 |
Oral GLP-1 gene delivery by neonatal Fc receptor mediated transportation of Fc fused nanocarrier as noble therapeutic for type 2 diabetes treatment. |
초록 |
Type 2 diabetes results from the ineffective use of insulin in the body. GLP-1 is a 30 amino acid peptide, stimulates insulin secretion and suppresses glucagon secretion thereby lowering blood glucose level. GLP-1 secretion is decreased in diabetic patients and half life time of GLP-1 is only ~2mins. We designed a IgG-Fc conjugated nanocarrier with protamine which forms a stable complex with GLP-1 gene and protects it from acid and enzyme degradation in the body. Fc has a longer life time and can increase the life time of GLP-1 by FcRn recycling pathway. The carrier has a Sulfo-SMCC linker that links Protamine-GLP1 complex and cystein modified Fc domain. The morphology of GLP-1 gene complexed with Protamine-SMCC-Fc was investigated by SEM and AFM. Higher cellular uptake by FcRn positive HT-29 cells confirmed the effective Fc interaction with FcRn. We hypothesized that the designed nanocarrier can be an excellent tool for successful oral delivery of GLP-1 and treat type 2 diabetes. |
