제목 In Situ Forming Hydrogel Encapsulating Upconversion Nanoparticles for Wireless-Optogenetic Neuronal Therapy
초록 Optogenetics is a technology using light-sensitive opsins for manipulating neuron circuits. In this work, optogenetic system with in situ hyaluronate hydrogel tethering upconversion nanoparticles (UCNP) was developed for non-invasive control of brain neural signal. Because UCNP emits visible light after absorption of skin penetrating near-infrared (NIR) light, they were used as a wireless light source. 980nm NIR laser is used to activate opsins, Channelrhodopsin (ChR2) and halorhodopsin which are transfected to the brain neuron. In situ hydrogel was prepared for long-term maintenance of UCNP near targeted neurons. The characteristics of UCNP were analyzed by fluorometer, DLS analysis and TEM imaging. ChR2 transfection was confirmed by YFP expression and the working of ChR2 function was observed from Ca2+ imaging. Taken together, we could confirm the feasibility of the wireless optogenetic system for a follow-up research about treatment of neurological disorders.
