제목 Optimization of Culture Conditions by Response Surface Methodology for Amylase Production by Arthrobacter sp.
초록 In this study, the physical factors for the production of amylase by Arthrobacter sp. were optimized using the reaction surface methodology (RSM). Physical factors including various inoculum volume(0.318-3.682ml), pH(5.318-8.682) and medium volume (16.364-83.363ml) were examined to screening for enhanced amylase production. Amylase activity was as low as 1.66mU/L before optimization after flask culture. Predicted results based on RSM, the optimal condition for distilled production amylase, were 2.84mU/L of estimated amylase production at 2.49mL inoculum volume, pH 6.85 pH and 42.87mL medium volume. Experimental amylase activity was 2.50mU/L, which was increased by 150% as compared with that before the optimization.
