Increasing contrast intensity around cancer region is an essential to determine accurate diagnosis and extent of resection. However, there has been a tendency to focus on stability as well as contrast intensity of contrast agents (CAs). There are numerous reports of free gadolinium-induced Nephrogenic system fibrosis in kidney patients. To prevent this, it is necessary to develop a CA using a stable macrocyclic chelator. In this study, we designed T1 CA using DOTA for stable contrast. DOTA was conjugated with saccharide to improve the water proton density by hydrogen bonding, 1H & 13C NMR analysis were conducted to demonstrate the synthesis of CA. in addition, xylenol orange method and ICP-MS were conducted to demonstrate Gd3+ tightly bind to CA. In vitro & In vivo paramagnetic properties of CA showed relatively high intensity compared to conventional CA due to high water proton density. These results demonstrated that our CA can be used as a cancer diagnostic CA. |
