제목 Chitosan-lipoic acid graft copolymer as a novel carrier for drug delivery
초록 Chitosan is produced by the deacetylation of chitin. CH is a copolymer of β(1→4) linked glucosamine and N-acetyl glucosamine. The advantages of CH are the biocompatibility, biodegradability. Lipoic acid is a naturally occurring dithiol compound. LA has been described as the potent biological antioxidant, the detoxification agent. Hydrophilic CH was synthesized with hydrophobic LA to form amphiphilic copolymer. This copolymer could form self-assembled nanoparticles in aqueous solution. Their sizes were ca.300nm measured by TEM and DLS. Fluorouracil has been widely used in the therapy of different solid tumor types such as cancer of the stomach, liver, and so on. However, 5-FU may cause some side effects. Hence, we tried to develop the novel drug carrier using CH and LA. 5-FU could be loaded in the hydrophobic core of nanoparticles. The loading efficiency and the release profiles of the CH-LA nanoparticles were measured by HPLC. Their loading efficiencies were more than 70%.
