For the production of bio-fuel, many researches have been actively conducted on discovering and improving the excellent microalgae species capable of high-speed growth and high lipid content. In order to create commercially valuable microalgae, it is essential to have method putting a superior trait DNA into cells. In many cases, exogenous materials delivery is highly dependent on the cell cycle status and light dependent growth of C. Reinhardtii. In mid-log phase growth, it facilitates maximum exogenous DNA uptake. However, It takes many time to prepare cells in mid-log phase. Therefore, it is necessary to find more efficient ways to reduce cell preparation time. One way to reduce cell preparation time is increasing initial concentration. Also, algal cultures can be synchronized naturally by alternating light/dark periods so that growth occurs in the light and DNA replication(s) and nuclear and cellular division(s) occur in the dark. Therefore, we also investigated the effects of this synchronized culture on the transformation too. |
