제목 Metabolic Engineering of microorganisms for the production of 3-hydroxypropionic acid from glycerol
초록 For every 9kg of biodiesel production, about 1kg of crude glycerol is generated as a byproduct. Glycerol is a good carbon source for the production of 3-hydroxypropionic acid (3-HP). Several engineered microbes such as Escherichia coli, Klebsiella pneumoniae and Pseudomonas denitrificans have been developed for the production of 3-HP by introducing an aldehyde dehydrogenase and a glycerol dehydratase. However, both the enzymes were seriously inactivated during the reaction. To minimize the inactivation of the enzymes, balanced expression of the genes was attempted by promoter engineering and unique dynamic promoters which are induced by 3-HP have been employed. The dynamic promoters could express the genes of the two enzymes continuously and maintained their activity until the end of fermentation. This talk summarizes our long efforts for the 3-HP production and discusses the potential and limitations of our work. Also, glycerol metabolisms affecting 3-HP production are discussed.
