제목 Novel injectable Tetronic-PLA-heparin conjugate for protein-based signaling system
초록 The use of biodegradable polymers has recently been proposed for several applications including tissue engineering, , agene therapynd novel drug delivery systems. The interaction of heparin with specific proteins (growth factors and cytokines) facilitates a wide range of fundamental biological processes including cellular proliferation, differentiation and tissue homeostasis. The incorporation of heparin to biomaterials has been widely studied to improve the biocompatibility (blood and cell) of materials. The objective of the present study is to investigate the growth factor release profile and in vitro cell compatibility of PLA-heparin conjugate using thermosensitive multifunctional Tetronic for pharmaceutical and biomedical applications as an injectable material for tissue regeneration and drug delivery. Tetronic-PLA copolymer was synthesized by ring opening polymerization of L-lactide with Tetronic using stannous octoate. Then, heparin was coupled to Tetronic-PLA copolymer by 1-ethyl-3-(3-dimethylamino propyl)-carbodiimide (EDC) and N-hydroxysuccinimide (NHS). The synthesized Tetronic-PLA-heparin conjugate was characterized by FT-IR, NMR, GPC, DLS and colorimetric assay. The thermo-responsive behaviors of aqueous polymer solution were investigated by vial tilting method. In-vitro bFGF release profiles and cell studies showed nontoxic and biocompatible properties. These results suggest that Tetronic-PLA-heparin conjugate would be very useful in biomedical applications as an injectable material for tissue regeneration.
