The development of metabolically engineered E. coli strains harboring a heterologous PHA synthesis and degradation pathway, and a novel strategy for the production of enantiomerically pure R3HB by these strains was demonstrated. R3HB can be efficiently produced by employing metabolically engineered E. coli harboring genes encoding the enzymes relating for biosynthesis and degradation of PHA. By using two-plasmids system, R3HB was most efficiently produced with 79.3% of yield from glucose. Even though, the product was mainly in the form of dimer, it can be easily hydrolyzed to monomer by alkaline heat treatment. Especially, the rate control of PHA biosynthesis and depolymerization can be a key for more improvement of the production efficiency. Furthermore, it is also promising to achieve further improvement of the R3HB production and the monomer fraction by controlling the expression levels of the related genes. |
