A fairly new approach in vaccine research is the development of antigen-encoding plasmid DNA as immunogen. To deliver DNA vaccine into the antigen presenting cells (APCs), such as dendritic cells and macrophages, non-viral vectors are strongly in need as safer alternatives to viral vectors. In this study, to induce the receptor-mediated endocytosis into APCs having mannose receptors, mannosylated chitosan(MC)/DNA complexes were prepared as gene carriers. Chitosan was coupled with mannopyranosylphenylisothiocyanate bearing mannose group and the composition of MC was determined by NMR spectroscopy. Complexes were induced to self-assemble in distilled water by mixing DNA with appropriate polymer solution at the desired charge ratios. Complex formation and DNAseI resistance were confirmed by electrophoresis and particle sizes and morphology were determined using TEM. Cytotoxicity and transfection efficiency of MC will be reported.
