제목 Attenuated responses& behaviors of vascular smooth muscle cells onto catechin-eluting stent coatings and action mechanisms of eluted catechin
초록 The suppressive effects of epigallocatechin gallate (EGCG) eluted from poly(lactide-co-ε-caprolactone, PLCL) stent coatings were investigated on the responses and behaviors of vascular smooth muscle cells (VSMCs). The involvement of nuclear factor-κB (NF-κB) was also examined as the mechanisms of EGCG-mediated inhibition. The proliferation and migration of VSMCs onto EGCG-eluting PLCL were significantly inhibited. VSMC cultures onto EGCG-eluting PLCL resulted in induction of cell cycle arrest at G0/G1 phase, leading to subsequent apoptosis. Expression of active matrix metalloproteinase-2 was directly lowered by EGCG and indirectly inhibited by EGCG-mediated suppression of NF-κB. Here were found the incorporation of FITC-conjugated EGCG into the cytosol of VSMCs and its further nuclear translocation. Taken together, the attenuated responses of VSMCs onto EGCG-eluting stent coatings was shown to be mediated through the suppression of NF-κB by EGCG incorporated into the cells.
