제목 Nonviral genome editing based on a polymer-derivatized CRISPR nanocomplex for targeting multidrug-resistant bacteria
초록 The dramatic increase in multidrug resistance due to the overuse of antibiotics has brought out the need for advanced therapeutics with narrow spectrum. In this study, we report a non-viral genome engineering system that can molecularly target these superbugs in narrow spectrum. We apply the typeⅡ CRISPR system into a nonviral nanocomplex, by covalent modification of Cas9 and subsequent complexation with sgRNA. We show that the CRISPR nanocomplex targeting mecA - the gene involved in methicillin resistance - can be proficiently delivered into Methicillin-resistant Staphylococcus aureus (MRSA). The CRISPR nanocomplex proved sufficient functional activity  which was comparative to native Cas9. The present study shows the prospective applicability of the CRISPR nanocomplex in the clinic as a target specific antimicrobial and can ultimately contribute to disease control and prevention.
