Synthetic biology is the study to design and construct new biological parts, devices and systems or re-design natural biological systems for useful purposes. When using genetic devices for a variety of applications, the operational range and dynamic range should be adjusted in the dose-response curve to suit the requirements. Therefore, we constructed riboswitch-based genetic devices with fast and easy methods without any information such as the sequences and structural information of riboswitches. Genetic circuits with a coenzyme B12 riboswitch were constructed for modular tuning of existing riboswitches. Two transcriptional repressor sets were used to compose the genetic circuits. Moreover, tuning the circuits performed with various strength of constitutive promoters to regulate expression of riboswitch and the repressor gene. As a result, the circuits composed of different repressor sets and promoter strength show different dose-response curves. |
