제목 Effect of homologous overexpression of citrate synthase on glyoxylate pathway genes and acetate assimilation in Pseudomonas putida
초록 Glyoxylate pathway plays important role in the assimilation of acetate for growth in various microorganisms including Pseudomonas putida. Once acetate is converted into acetyl-CoA, percentage of Acetyl-CoA funnelling between to TCA or glyoxylate cycle and PHA synthesis is not explored. In this study, we homologously overexpressed citrate synthase gene to divert acetyl-CoA to TCA/glyoxylate cycle. The effect of citrate synthase overexpression on other glyoxylate pathway genes was quantified using RT-PCR and acetate metabolization was compared between wild type and mutant P. putida. This study provide an potential application of bioconversion of abundant but refractory simple acetic acid into useful metabolites and chemicals which dependent on glyoxylate cycle.
