Guided Neurite Outgrowth through Porous Matrix with NGF Gradient |

The sufficient functional recovery of damaged peripheral nerves has been considered as one of the clinical challenges. A therapeutic aim after peripheral nerve injury is thus to re-establish connectivity by aiding in axonal regeneration and correct axonal guidance. It is well known that the nerve regeneration in the body is mediated by gradients of bioactive molecules including nerve growth factor (NGF; chemotaxis). In this study, we prepared a porous polycaprolactone (PCL)/Pluronic F127 membrane with NGF gradient along the designated direction. The NGF loading amount, NGF release profile at different positions along the NGF gradient direction of the prepared membrane, and neurite outgrowth behavior of PC12 cells on the NGF gradient membrane were investigated (This work was supported by a grant from the National Research Foundation of Korea (grant No. 2011-0000244)). |
