Immobilization of recombinant Salmonella typhimurium TA1535 as a biosensor |

Genetically engineered Salmonella typhimurium TA1535 cells were immobilized to 96well plate as a biosensor. Salmonella typhimurium TA1535 cells contain an inducible SOS promoter that controlled promoterless luxCDABFE genes from Photobacterium leiognathi as reporter. Therefore, bioluminescence occurs as a response to the presence of DNA-damaging agents that stimulated SOS promoter. For implementation result of Salmonella typhimurium TA1535 as a biosensor, optical density and cell concentration have been optimized to be OD=0.8 and 1010 CFU/ml. In this experiment, bacterial cells were immobilized with agar and solidified in 96well plate to handle easily and to keep long time. Biosensor plates that had one, two, four and six week preservation periods were treated with a typical mutagenic chemical, mitomycin C. When kept at 4 ℃ with preventing evaporation, found to be the biosensor shows quite reasonable and reproducible results until 6 weeks. In X-ray and UV exposure experiments, it was also found to be very convenient than using normal cell cultures. |
