제목 Synthesis and in vitro evaluation of injectable hyaluronic acid based hydrogel
초록 Acrylation of hyaluronic acid (HA) was achieved by two step synthesis protocols. Derivatized HA and thiolated poly(ethylene glycol) were crosslinked by Michael type addition reaction for the preparation of injectable hydrogels. The degree of acrylation of HA was determined to be around 20% at the physiological condition. Gelation kinetics was shown that hydrogel was formed within 20 minutes. Mechanical properties of hydrogels was elucidated by varying concentrations of acrylated HA and thiolated poly(ethylene glycol) ;1:1, 1:2, and 1:3. It has been shown that gelation kinetics, mechanical properties, swelling ratio and degradation rate was affected by the ratio of HA and PEG. Human mesenchymal stem cells are cultured in the HA based hydrogel in vitro. Proliferation of human mesenchymal stem cells (hMSCs) was examined in different hydrogels. Cellular proliferation rate is dependent on the composition of hydrogels. The effect of BMP to the hMCSs was also evaluated by adding BMP into the hydrogel with hMSCs. The possibility of guiding differentiation of hMSCs in the gels was also shown in vitro. HA based injectable hydrogel has a great potential as a scaffold system and can be used for cell delivery system as well as growth factors for facilitating neo tissue generation in vivo.
