Imbalance between glycolysis and product synthesis may accumulate pyruvate or reduce the productivity. Specifically, the amount of pyruvate can determine the production condition. Therefore, balanced maximization of the pathway efficiency between glycolysis and product-forming pathway is required to construct the optimal microbial cell factory. We propose synthetic UTR design of ptsG which effectively controls overall glycolytic flux as a metabolic valve. Using the metabolic valve, optimal glycolytic flux was explored to maximize the production of model system. Tuning down of glycolytic flux significantly reduced the pyruvate leakage, which increased 20% in yield. Amplification of glycolytic flux led to the 7% and 12.5% higher productivity of butyric acid and 2, 3-butanediol, respectively. Our results demonstrated that fine control of the activity of PtsG allows the effective metabolic valve to accomplish the maximum metabolic performance. |
