A variety of tissue engineering approaches are motivated by the clinical need for articular repair. Alginate has been used as a biomaterial for cartilage regeneration. Purified alginate microcapsules is fabricated by seed cartilage cell into microcapsules. After 1, 3 , 7 day of cell seeding, cell proliferation activity was measured via MTT assay. Morphological observation, histology, biological assay for collagen type Ⅰ, Ⅱ and aggrecan, and PCR were performed at each time point 1, 2 , 3 weeks. In result, when we cultured cartilage cell of rabbit in vitro, we observed better to keep the characteristic of cartilage cell in the Purified alginate microcapsules than Non-purified alginate microcapsules. This study suggests that Purified alginate microcapsules may serve as a potential cell delivery vehicle and a structural basis for in vitro tissue engineered articular cartilage.
