제목 In vitro Test for the Bone Marrow Stromal Cells with Silk Fibroin Contents in Silk/PLGA Scaffold
초록 Silk Fibroin(SF) has been increasingly studied for new biomedical applications due to the biocompatibility, slow degradability and remarkable mechanical properties of the material. We examined the effects of silk fibroin scaffold properties on BMSCs attachment, proliferation and differentiation. We developed the SF loaded poly(L-lactide-co-glycolide)(PLGA) scaffolds. SF/PLGA 0, 10, 20, 40 and 80 wt% scaffolds were prepared by solvent casting/salt leaching method. These cells were seeded in prepared SF/PLGA scaffold. The effect of SF/PLGA scaffold on the proliferation and differentiation of rat BMSCs were assessed in culture using the MTT assay and RT-PCR was conducted to confirm mRNA expression of ALP and Osteocalcin. According to our results, 80% SF/PLGA scaffold have good cell compatibility and attachment was higher. This Research was Supported WCU (R31-20029) and by SCRC (SC4110).
