Stent surface modifications for strong binding of cell-derived extracellular matrix toward outgrowth endothelial cells delivery |

In previous study, cobalt-chromium (Co-Cr) surface was modified by introducing a self-assembled monolayer of phosphonic acid (PA-SAM) and fibroblast-derived extracellular matrix (FDM) to deliver outgrowth endothelial cells (OECs). FDM was harvested by decellularizing fibroblasts cultured on the culture plate and was grafted on the PA-SAM-coated Co-Cr. The current study is focused on directly binding FDM on a modified Co-Cr for giving more natural extracellular environment. First, polydopamine (pDA) and fibronectin (FN) were in turn introduced on Co-Cr. It is known that pDA is a strong adhesive molecule and FN robustly binds to ECM components through several binding domains. Herein, FN layer was also strongly bound to the pDA-coated Co-Cr. The tightly immobilized FN is not only suitable for the culture of fibroblasts but also plays a role in holding ECM left after decellularization. This strategy can be a promising method for the formation of a durable OEC layer on a coronary stent. |
