Since stem cells have the potential to differentiate into specialized cell types under appropriate conditions, tremendous efforts have been given to differentiate stem cells into specific cell lineages, such as osteoblast, chondrocytes, adipocytes, and myoblasts. In this study, adipose-derived stem cells(ASC) were isolated and then subjected to chondrogenic differentiation using biodegradable, peptide-grafted poly (L-lactic acid) (PLLA) scaffolds. Prior to cell seeding, the scaffolds were activated with plasma treatment to introduce hydrophilic surface and reacted with Gly-Arg-Gly-Asp (GRGD) to promote cell adhesion onto the substrate. Cultured in the chondrogenic-defined medium for specific time, chondrogenic differentiation of ASC were evaluated using WST-1 assay, histology, western blotting, and gene expression of collagen type I, II, X, aggrecan, and Sox 9. |
