제목 | Chitosan Microspheres for a Nasal Vaccine Delivery System in vitro and in vivo |
초록 | Chitosan is a valuable excipient for nasal drug delivery system due to its biocompatibility, biodegradability and low cost. Also, it has been reported that chitosan could induce cytotoxic T lymphocytes and macrophage activation. Chitosan microspheres (CMs) were prepared based on ionic gelation process of tripolyphosphate and then, the microspheres were loaded with Bordetella Bronchiseptica Dermonecrotoxin (BBD), a virulence factor of a causative agent of atrophic rhinitis, for nasal vaccination. Particle sizes of BBD-loaded CMs were smaller than 10 μm and had narrow size distribution with uniformity. Vaccine BBD release from BBD-loaded microspheres and secretion of cytokine such as tumor necrosis factor-α (TNFα) and nitric oxide (NO) from BBD-loaded CMs were checked in vitro (Fig. 1). Also, BBD-loaded CMs were treated to mouse as a model animal, and systemic and nasal immunities were checked in vivo (Fig. 2). Fig. 1.
