제목 Effect of Acyl-CoA’s on the Production of Doxorubicin in Streptomyces peucetius ATCC 27952
초록 An anticancer agent doxorubicin biosynthesis is initiated from one propionyl-CoA starter unit and nine malonyl-CoA extender units in S. peucetius. We increased the acyl-CoA pools by integrating E. coli pantothenate kinase gene into S. peucetius to produce more doxorubicin. Pantothenate kinase is the key regulatory enzyme in CoA biosynthetic pathway. As a result, ε-rhodomycinone, a major aglycon intermediate of doxorubicin, production was enhanced. To produce more doxorubicin from this ε-rhodomycinone, glycosyltransferase or glycon-biosynthetic genes were overexpressed in panK-integrated S. peucetius. Major metabolites from these strains were quantified and compared.
