제목 Reversal of adenosine-rich tumor microenvironment via antibody-modified oxygen nanosome
초록 Hypoxia, which is formed around tumor, acts as an immune checkpoint, making immunotherapy difficult. Hypoxic Tumor cells express surface markers such as CD39 and CD73 by the hypoxia inducer HIF-1a. They break down ATP outside the cell into adenosine. Adenosine induces immunosuppression by binding to GPCR present on various immune cells. Thus, oxygen-carrying oxygen nanosome were fabricated to reverse the hypoxia and prevent accumulation of adenosine.

Immunonanosome synthesized by modifying the surface of oxygen nanosome with CD73 antibody, enabling effective oxygen delivery to tumor cells in a hypoxia. Antibody modifications were confirmed through various characterization methods, and it was confirmed that it has no cytotoxicity. It was confirmed through HPLC and adenosine assay that the amount of adenosine in the TME decreased as the immunonanosome was treated on the hypoxic tumor cells. In addition, the decrease in the surface expression rate of CD73 was confirmed through immunostaining as the hypoxic tumor cells were treated with immunonanosome. Futhermore, as adenosine in TME decreased, activation of immune cells was confirmed through ELISA.
