Oxygen supply enhanced microwell arrays for 3D cell culture system |

Engineered 3D cell cultivation systems for the production of multi-cellular spheroids have attracted considerable attention. However, in recent advancement of microwell arrays for tissue engineering applications, oxygen delivery to spheroids has not been fully addressed, which made users suffer from the low viability and functionality of the reconstituted tissues. Here we develop noble networked concave microwell arrays for the effective construction of 3D spheroids. We also reveal that the oxygen distribution around the spheroids is finely controlled by varying the bottom thickness of the gas permeable concave microwell. Our results demonstrate that the multi-cellular functionality can be tuned up by networking individual 3D spheroids without additional chemicals or biological supplements. We anticipate our results to be used in high-throughput screening platforms to study cell-cell interactions and in the development of customized 3D tissue culture systems. |
