Recently, novel DNA oligomer with reactive function was designed by incorporation of oxanine (Oxa) into DNA oligomer. Since Oxa has O-acylisourea structure in its base-ring, covalent amide-bond is formed with amine-containing molecules without any chemical activation. By employing Oxa-DNA on the amine-functionalized materials (chip-surface or nano-particle), DNA-immobilized systems were constructed efficiently. Interestingly, Oxa-used linkage system was found to show high immobilization and hybridization efficiencies compared to chemical-used linkage system. In addition, when Oxa-DNA was combined with DNA-binding protein, novel DNA-protein crosslink complex was designed. Such bioactive DNA-protein crosslink was efficiently used as bioprobe molecule for analyzing molecular interaction mechanisms. The developed reactive DNA oligomer and bioactive DNA-protein crosslink will be used as new functional biomolecules for development of novel bio-medicine and nano-biotech system. |
