Biophysical Investigation of Neuronal Outgrowth Kinetics in Artificial Three-Dimensional Neuronal Networks |

We report that the neuronal outgrowth kinetics in 3D neuronal networks could be effectively modulated by controlling the network geometric factors such as bead-inclusion, density, and diameter of electrospun polystyrene (PS) fibers. The PS fibrous network with microsize beads formed by unstable electrostatic force afforded structural heterogeneity so that the neuronal cells could settle down on the beads inside of PS web and subsequently extend z-directional axonal outgrowth along PS fibrous structures. It was shown that the growth cone migration rate and axonal steering were strongly influenced by PS fiber density and that there was a strong correlation between fiber diameter and focal adhesion protein expression. |
