Efficient microRNA Detection with Highly Magnetized Encoded Hydrogel Microparticles |

Polyethylene glycol-based microparticles garnered considerable attention on microRNA detection owing to their solution-like reaction environment thus eventually leading to high sensitivity and specificity. However, several particle rinsing steps in the assay procedure present challenges for rapid and efficient detection. To overcome this limitation, we encapsulated magnetic nanoparticles with high density to reduce the rinsing steps and duration via magnetic separation. A large number of magnetic nanoparticles were encapsulated into hydrogel microparticles based on discontinuous dewetting technique combined with degassed micromolding lithography. To figure out the assay performances of our particles, we attached DNA probes to the particles and detected three preeclampsia-related microRNA targets. The particle rinsing durations were decreased by a factor of 3.8 owing to magnetic force while we were able to keep the advantages of nonmagnetic particles in terms of assay performances. To demonstrate multiplex availability, we detected the said targets simultaneously. We observed similar levels of fluorescence intensity and no significant cross reactivity between each target. |
