제목 | The reversal effect of hypoxic condition for cell via Oxygen nanobubble(ONB) |
초록 | When cancer develops, the surrounding tissues become deficient in oxygen supply. Hypoxia increases the expression of HIF-1a due to a lack of oxygen supply. Thus, a few membrane proteins act as an efflux pump that release the drugs out of the cell. It lowers the drug efficiency. So we fabricated an oxygen nanobubble(ONB) consisting of a phospholipid shell and an oxygen core to reverse hypoxia. Cellular morphology and oxygen distribution were changed in hypoxia. Cells under hypoxic condition didn’t grow well and became small-sized spherical shape. We fabricated ONB and confirmed ONB has no toxicity through the cell cytotoxicity test. The effect of ONB to helps reverse the hypoxia was confirmed by image-IT hypoxia reagent and live/dead assay. With ONB treatment, the number of hypoxia cells were decreased. And increased expression of HIF-1a in hypoxia cells was confirmed using ELISA. It is expected that ONB can be used for oxygen delivery to cells in the core of spheroid and organoid. |
