Molecular Imaging of Drug-Induced Tumor Apoptosis with a Caspase-3/7 Triggered Nanoaggregation Probe |

In this work, we have developed 18F-labeled and Cy5.5-labeled caspase-3/7 sensitive nanoaggregation probes for in vivo imaging of apoptosis. These probes contain a peptide substrate (Asp-Glu-Val-Asp) which can be cleaved by caspase-3/7. Once it is cleaved by the enzyme in the apoptotic cell, the remaining part undergoes intramolecular cyclization that provides hydrophobic macrocycles. Self-assembly of cyclic monomers can form a large particles by the hydrophobic interaction that results in trapping and enhanced retention of the signal in the apoptotic tumors. Results will be presented that synthesis and labeling procedures of the probes, in vitro evaluation of the labeled probes and specific apoptosis imaging with doxorubicin-treated tumor xenografts.
