Synergy of methylerythritol phosphate pathway and a short-cut route to 1-deoxy-D-xylulose 5-phosphate for enhancement of sesquiterpenoids production from methane in a methanotrophic bacteria |

Sesquiterpenoids belong to the largest subgroup of isoprenoids exhibit many attractive features in industrial applications. Recently, methanotrophs have been considered as promising platforms for the production of valuable products from a cheap C1 substrate, methane. Taking advantage of the ribulose 5-phosphate pool in Methylotuvimicrobium alcaliphilum 20Z, we here employ a short-cut route from this intermediate to 1-deoxy-d-xylulose 5-phosphate (DXP) in combination with (MEP) pathway to efficiently improve isoprenoids production. Together with that, the fusion of nDXP enzyme and DXP reductase was employed increasing the α-humulene production yield up to 2.8-fold. These engineering strategies also enhanced the titer of another sesquiterpenoid, α-bisabolene, up to 2-fold higher than that of the prior strain. This study demonstrated the potential of the synergy of MEP and nDXP pathways for enhancing the production of various sesquiterpenoids from methane in type I methanotroph. |
