제목 | Oxygen nanobubbles(ONB) for the reversal of hypoxic cells in microenvironments |
초록 | When cancer develops, tissues are deficient in oxygen. Hypoxia increases resistance to drugs, making treatment difficult. We fabricated an ONB consisting of a phospholipid shell and an oxygen core. it is non-toxic to cells and helps reverse the hypoxia. Cellular morphologies and oxygen distribution was changed by the hypoxia in different cells(e.g. cancer cells, normal cells, and macrophages). Cells grown under hypoxia conditions didn’t die but did not grow well either, and they were spherical in shape. This was confirmed by microscopy and live/dead assay. The oxygen distribution was confirmed by image-IT hypoxia reagent. Cells also express HIF-1a due to a lack of oxygen. Increased expression of HIF-1a in hypoxia cells was confirmed using ELISA. When ONB is treated on these hypoxia cells. The cells begin to grow again, and the oxygen distribution increases. It is expected that oxygen nanobubbles can be used for oxygen transfer of cells in the production of spheroids and organoids. |
