제목 A NADH dependent lactate dehydrogenase from Alcaligenes eutrophus H16 reduces 2-oxoadipate to 2-hydroxyadipate
초록 Adipic acid is used in the production of nylon-6,6 polyamide. Currently it is produced only from non-renewable sources by chemical methods. Its production by recombinant microorganisms is often limited by capability of pathways. Adipic acid can be produced by combining lysine production and glutamate degradation pathways by reducing 2-oxoadipate to 2-hydroxyadipate, which remains a major hurdle. We report an enzyme lactate dehydrogenase from Alcaligenes eutrophus H16 that efficiently reduces 2-oxoadipate to 2-hydroxyadipate and bridges these two pathways. The identified gene ae-ldh was cloned, expressed in Escherichia coli, purified, and characterized for its properties. The recombinant enzyme with a molecular weight of 36.7 kDa exhibited broad substrate specificity for various 2-oxoacids and showed 50 % activity on 2-oxoadipate than pyruvate, natural substrate. NADH was the preferred coenzyme for the reduction of most 2-oxoacids tested.
