Adaptive laboratory evolution (ALE) allows adaptive changes in microbial populations over a long period of time. ALE is often used as a metabolic engineering strategy to improve the target production by allowing cell population to adapt to culture condition. ALE is especially useful when no clear metabolic engineering target remains. One problem with the current ALE methodology is that it takes a relatively long time to obtain a positively evolved strain. To address this issue, we devised a genetic evolver device that consists of synthetic gene circuit and various mutator combinations. The genetic evolver devices accelerated the mutation frequency in a tightly controlled manner. Each mutant combination showed different mutation rates and various basal expression levels. Using the genetic devices, we will be able to improve mutagenesis rates during adaptive laboratory evolution, leading to the faster engineering of the target cells. |
