Cell-free synthesis and analysis of enzymes having industrial implications |
초록 |
While microbial cells have widely been used for the production of recombinant enzymes, the use of living cells sets intrinsic limitations in numerous dimensions including the throughput of process and choice of assay substrates. In addition, intracellullar conditions are often incompatible with proper expression and folding of target proteins. These limitations associated with cell-based expression can be effectively overcome by employing cell-free protein synthesis as an alternative expression platform. Not being subject to the constraints of maintaining cell viability and intact cell membranes, cell-free protein synthesis offers greater flexibility in designing expression and screening procedures. In this study, we will discuss different approaches to harness the advantages of cell-free synthesis for efficient production of enzymes that have industrial implications in different sectors of industrial biotechnology. |
