제목 Classification of oxidative damage chemicals using stress-specific bacterial biosensors
초록 The characterized redox-sensing promoters from SoxRS and OxyR in Escherichia coli that regulate the responses to oxidative stress have been characterized. We have constructed a recombinant bioluminescent Escherichia coli strain, EBJM1 (pqi-5::luxCDABE), containing a promoter for unknown function (pqi-5) gene a member of soxRS regulon, fused to the Vibrio fisheri luxCDABE operon. The soxRS-dependent induction of pqi-5 occurred by paraquat, a superoxide radical-generating agent. Different patterns of stresses response sensitivities have been compared for some oxidative stress-chemicals, such as hydrogen peroxide, cadmium chloride, and paraquat, with EBJM1 including existing oxidative stress responsive strains, EBHJ1 (sodA::luxCDABE), which is strongly induced by redox-cycling agents, and DPD2511 (katG::luxCDABE), which is sensitive to hydrogen peroxide. It was found that the responses from the pqi-5-, sodA- and katG-based strains were significantly different dependent upon the chemicals being tested. The classification of these chemicals by three different groups, soxRS-response group, oxyR-response group, and etc. group was possible.
