Micrometer sized 4-cyano-4´-pentylbiphenyl (5CB) droplets were functionalized with Poly(acrylicacid-b-4-cynobiphenyl-4´-undecylacrylate) (PAA-b-LCP) using microfluidics technique. The LCP block was strongly anchored in the 5CB while the PAA block formed a brush like structure around the droplets (5CBPAA). The 5CBPAA were used for the immobization of enzyme such as glucose oxidase (5CBPAA-GOx) and urease (5CBPAA-urease), and biotinylated (5CBPAA-biotin) with the aid of chemical coupling agents. The 5CBPAA-GOx, 5CBPAA-urease, and 5CBPAA-biotin droplets were tested for glucose, urea, and biotin-avidin complexation, respectively, under the tested optimized condition of concentration and pH. The detection of the glucose, urea and avidin-biotin complexation were successfully characterized by configurational change of the 5CB droplets such as a radial-to-bipolar change in glucose and avidin, and a bipolar-to-radial change in urea solutions observed via a polarized optical microscope at 5CB/aqueous interface. These functionalized droplets were having high sensitivity, specificity, and stability to their respective analyte, which demonstrate the versatility of 5CBPAA droplets that can make selective detection by immobilization of selective enzymes or through a ligand/receptor model. |
