Polymorph Screening Technology: Molecular recognition perspective of solvent effect on the crystallization of active pharmaceutical ingredients(APIs) |

This contribution reports the effects of molecular recognition events in the crystallization of the polymorphic forms of clopidogrel hydrogen sulfate (CHS). Solvents were chosen on the basis of their power of dissolution of the API, the solubility parameter and hydrogen-bonding potential. Experiments were conducted to (1) measure solubility and induction times, (2) monitor solution concentrations and solid phase compositions with in-situ ultrasonic velocity for seeded and unseeded systems, and (3) identify intermolecular interactions between solvents and CHS molecules. Results show that relative nucleation rates of CHS polymorphs are dependent on the hydrogen-bonding nature of the solvent and are independent on solubility. It is concluded that intermolecular interactions and specifically the hydrogen-bonding propensity of solvents with CHS molecules have profound effects on the selective crystallization of CHS polymorphs. |
