The aim of this study was to deliver gene and peptide for synergetic action against tumor. The ternary complexes of gene, peptide, and targeting peptide conjugated-LMWSC were prepared by electrostatic interaction in aqueous solution. The binding affinity, pDNA release, and stability from DNase of ternary complexes were confirmed by electrophoresis. The particle size and zeta potential of ternary complexes was evaluated by DLS. The result of tranfection assay showed that ternary complexes with targeting peptide conjugated-LMWSC was more effective than that without transferrin. The cytotoxicity and antitumor activity of ternary complexes were confirmed by MTT assay in normal and cancer cells. These results suggest that co-delivered gene and peptide can effectively inhibit tumor growth via receptor mediated endocytosis.
