Isolation of renal cells from adult rat kidneys and the effect of layer-by-layer assembly nanocoating on the proliferation and differentiation of renal progenitor cells |

Layer-by-layer self-assembly is a thin film fabrication technique that involves the deposition of alternating layers of oppositely charged biocompatible materials (polycation and polyanion). This technique offers physiological versatility by nanocoating of a single-cell to create a defined microenvironment for cell proliferation and differentiation. In this study we report isolation of progenitor cells from adult rat kidneys and its encapsulation using a layer-by-layer self-assembly technique with polyelectrolytes chitosan/ gelatin and alginate. Analysis of the nanoencapsulated renal progenitor cells showed that the layer-by-layer assembly did not affect the viability, and proliferation and differentiation of the cells for 21 days under 2D culture. Results showed that the single-cell encapsulation of renal progenitor cells was able to protect the cells without interfering with its biological activity and has potential for use in cell delivery where physiological versatility is necessary including, but not limited to the recellularization of acellular tissue constructs. |
