Linker-free antibody conjugation for hydrogel microparticle-based multiplex immunoassay |

As demands for detecting multiple proteins in a single sample have increased, encoded hydrogel microparticle-based multiplex immunoassay have gained much attention because of its high sensitivity and multiplex capacity rather than the enzyme-linked immunosorbent assay(ELISA). Previously, theseparticles were functionalized with linker(SH-PEG-NHS)-conjugated antibody after particle synthesis via stop flow lithography. However, the linker can damage antigen-binding fragment(Fab) region in antibodies and is vulnerable to hydrolysis in aqueous solution, which limits assay capability. Herein, we present a multiplex immunoassay platform that uses linker-free antibody, or half antibody produced by reduction of whole antibodies which has free thiols that can bond with double bonds in microparticles directly. |
