Oral delivery of therapeutic biomolecule has emerged as a promising field in the treatment of disease. In this study, we developed oral gene delivery system of glucagon-like-peptide-1 (GLP-1) to cure high-fat diet-induced diabetes. The GLP-1 gene was shielded with bPEI via charge-to-charge interaction and further wrapped with heparin-taurocholate bioconjugate (HTCA-PP). In addition, when the GLP-1/HTCA-PP complex was orally administered every 4 days for one month, the blood glucose levels were stably normalized within 150 mg/dL and body weight of all mice was significantly reduced. In the case of untreated mice, their blood glucose levels did not return to normal levels and their body weight was gradually increased. From the immunohistolotical analysis, GLP-1 was observed around pancreatic islets and in the small intestine. Our oral gene delivery system of GLP-1 gene combined with HTCA-PP non-viral vectors demonstrated a promising therapeutic potential to cure type 2 diabetes mellitus. |
