제목 Engineered 3D Lung Cancer Models to Explore Drug Resistance Mechanisms
초록 Understanding the relationship between tumor microenvironments and drug response is critical in developing cancer therapy. To accurately predict the drug response of cancer cells, it is necessary to mimic the properties of the native tumor microenvironments. Hydrogel-based 3D models have been considered as a promising platform due to their bioactivities and tunable properties. Herein, we develop an engineered 3D lung cancer model using gelatin and poly (ethylene glycol)-based interpenetrating network (IPN) hydrogels formed via horseradish peroxidase (HRP)-mediated oxidative cross-linking reactions. The 3D model is created by encapsulating human non-small cell lung cancer (NSCLC) cells within the matrices. Using the 3D tumor construct with similar mechanical strength to lung tissue, we investigate the drug resistance mechanisms against Gefitinib. In conclusion, our tumor model has great potential as a 3D platform to study basic cancer biology and explore drug resistance mechanisms.
