제목 | Antibiotic-loaded PLGA Particle for Local Delivery System |
초록 | Poly(D,L-lactid-co-glycolide) (PLGA) is biocompatible and biodegradable polymer used a wide range of controlled drug delivery application. Gentamicin sulfate (GS) is a higly water soluble antibiotic of the aminoglycoside group and widely used for the treatment of osteomyelitis and the prevention of secondary infection after orthopedic surgery. The mixture of GS with PLGA was melt extruded by mini max molder. The extrusion was grinded by freezer mill. The objective of this study was to prepare the biodegradable particle for sustained release of GS and PLGA mixture. The effects of particle size was investigated. In vitro study was performed in pH 7.4 phosphate buffered saline at 37℃. The morphology was observed by scanning electron microscope (SEM). Drug content and release amount of GS were analyzed by high performance liquid chromatogaphy (HPLC) and the released GS activity was observed by microbiological assay using Staphylococcus aureus.
